TScan Advances TSC-101; Phase 3 Topline Data Expected In 2028

(RTTNews) - TScan Therapeutics, Inc. (TCRX), a clinical-stage biotechnology company developing TCR-engineered T cell (TCR-T) therapies for cancer, has reported its second-quarter 2026 financial results and provided business updates, highlighting the Phase 3 advancement of TSC-101, encouraging Phase 1 ALLOHA data, commercial-ready manufacturing progress and expansion of its hematologic malignancy pipeline.

Recent Clinical & Corporate Updates:

-- A Phase 3 trial, evaluating TSC-101 in patients with hematologic malignancies undergoing allogeneic hematopoietic cell transplantation, dubbed ALLOHA-2, is underway.

The company expects to complete enrollment and report topline ALLOHA-2 data in mid-2028.

-- A Phase 1 interventional umbrella study evaluating the feasibility, safety and preliminary efficacy of an escalating dose regimen of up to 2 doses of TSC-100 and TSC-101 in patients with AML, MDS, or ALL following hematopoietic cell transplantation, known as ALLOHA, is also ongoing.

The company reported positive initial data from Cohort C of the ongoing ALLOHA Phase 1 study, evaluating TSC-101 generated with the commercial-ready manufacturing process, in patients with heme malignancies undergoing allogeneic hematopoietic cell transplantation this June.

According to the study results, the commercial-ready process achieved an approximately 90% first-pass manufacturing success rate, with 17 of 19 manufacturing runs successful. Despite high-risk disease characteristics, 79% of patients achieved complete donor chimerism within approximately three weeks of their first TSC-101 infusion, while the therapy remained well tolerated.

Pipeline Progress:

The company plans to share updated Cohort C ALLOHA data in Q4 2026, followed by additional data with more than one year of follow-up in the first half of 2027.

Phase 1 studies of TSC-102-A01 and TSC-102-A03, designed to expand the hematologic malignancy program to additional HLA types, are expected to begin in Q4 2026, with initial data anticipated in 2027.

For solid tumors, TScan is developing in vivo TCR-T engineering approaches, with an IND filing targeted for the second half of 2027. The company is also developing potential therapies for HLA-B27-associated autoimmune disorders, including ankylosing spondylitis.

TScan has an agreement with Cellares to evaluate automated manufacturing platforms as a potentially scalable and cost-efficient path toward commercial production.

Q2 2026 Performance:

TScan reported Q2 revenue of $1.1 million, compared with $3.1 million in Q2 2025, primarily reflecting the timing of research activities under its Amgen collaboration.

R&D expenses declined to $23.4 million, compared with $32.6 million in the prior-year quarter, mainly due to lower laboratory, research material and contracted service costs.

G&A expenses were $8.1 million, compared with $9.1 million in Q2 2025.

TScan reported a net loss of $30.4 million, or $0.23 per share, compared with a net loss of $37.0 million, or $0.28 per share, in Q2 2025.

The Road Ahead:

The key near-term catalyst is continued advancement of the Phase 3 ALLOHA-2 trial evaluating TSC-101 for relapse prevention following allo-HCT.

TScan expects to initiate Phase 1 studies of TSC-102-A01 and TSC-102-A03 in Q4 2026, while additional ALLOHA data are expected through 2027.

The company is also advancing its solid tumor TCR-T platform, with an IND filing targeted for H2 2027.

Cash Position:

TScan held $100.2 million in cash and cash equivalents as of June 30, 2026, excluding $5.0 million of restricted cash.

The company expects its existing cash resources to fund its current operating plan into Q2 2027.
